I assume, the isolation cells (even in transparent mode - sure) cause the "huge" delay.
No!

Most of the delay you are seeing is digital delay within the logic, not analogue delays in the pads and routing. Four clocks of digital delay at 150MHz is 26.7ns, so that's most of your "about 30ns".

The analogue delays are quite small; I don't think values have been published yet for RP2350 but they are in the datasheet for RP2040 (see 5.5.3.6). The pad delays there, at 3.3V, are max 2.14ns output, 0.76ns input. Including other sources of analogue delay within the chip that adds up to max 5.59ns output, 4.95ns input. Those are worst-case values; typically they will be smaller.
