Ok, removed the Pi5 from the 3D printed case and found the Pimoroni bottom's connection at the top into the Pi5 board didn't have the plastic fully pushed down. It must have wiggled its way loose over 6 months of daily in and out of the backpack.
Pi5 now boots and is operable, I hope there's no problems with the NVMe SSD, I might have to have a 2nd backup one to avoid a delay with no desktop.
