For best efficiency while discharging the 2s3p will be most efficient as that has the least voltage drop for the buck convertor (but check the minimum voltage specs for battery and convertor).
Always be aware that many lithium technologies have a thermal runaway fire risk, use proper charging and discharging protection methods.
Ideally if you can afford the loss of capacity do not charge above 80% and discharge below 20%, this not only makes it safer but extends the life of the batteries to thousands of cycles.
Or use LiFePO4 or NiMH batteries instead which are a lot safer than Li-ion, Li-poly etc.
