byobu is a group of scripts build around the program tmux. You can read the scripts (less /usr/bin/byobu-enable) to see what they are intended to do.
I notice that byobu-launcher-install adds code to ~/.profile, which should run during shell startup, but only in login shells. In a desktop environment, whether a new terminal window counts as a login shell depends on the settings of the terminal program you are using.
